A Small-Molecule Enzyme Inhibitor, Not a Peptide

5-Amino-1MQ is a small synthetic molecule — a methylquinolinium — and it is worth being clear that it is not a peptide, despite being sold alongside them. It works by blocking a single enzyme: nicotinamide N-methyltransferase, usually shortened to NNMT.

That enzyme does something deceptively simple. It attaches a methyl group to nicotinamide, the form of vitamin B3 the body recycles into NAD+, the molecule every cell depends on to run its energy-producing reactions. Methylated nicotinamide can no longer be recycled and is excreted. The enzyme therefore diverts material away from NAD+ production.

Interest sharpened when NNMT was found to be present at high concentrations in the fat tissue of obese animals and people. A single enzyme that both consumes methyl groups and removes NAD+ precursors, and that is unusually abundant in a tissue central to metabolic disease, is a plausible point of intervention — and inhibiting it is the cleanest way to test whether that plausibility holds.

Why 5-Amino-1MQ Is Studied

  • Protecting the NAD+ salvage pathway: Cells recycle nicotinamide back into NAD+ continually. An enzyme that methylates nicotinamide takes it out of that loop, so inhibiting the enzyme is a way of asking how much the loop matters.

  • Methyl group accounting: Every methylation draws on the same limited pool of methyl donors. An enzyme consuming that pool in quantity competes with everything else that needs it, including the machinery regulating gene expression.

  • An enzyme concentrated in fat tissue: NNMT is markedly more abundant in the adipose tissue of obese models. Whether that is a cause or a consequence is a live question, and inhibition is the experiment that distinguishes them.

  • Inhibition rather than activation: Most compounds here switch something on. Removing an enzyme's activity is a different experimental logic, and often a more interpretable one, because the result shows what that enzyme was contributing.

How It Works — In Simple Terms

NAD+ appears in a vast number of the reactions that release energy from food, and cells cannot maintain it by synthesis from scratch alone — the cost would be prohibitive. Instead they recycle. NAD+ is consumed, nicotinamide is released, and nicotinamide is converted back into NAD+. This salvage loop is how the supply is sustained, and it runs constantly.

NNMT interrupts the loop at a single point. It attaches a methyl group to nicotinamide, producing a molecule the salvage pathway cannot use and the kidneys excrete. Each methylation therefore costs twice: one methyl group is spent, and one NAD+ precursor is lost. 5-Amino-1MQ occupies the enzyme's active site so the reaction cannot proceed.

Where methyl groups come from is the part usually left out. They are supplied by S-adenosylmethionine, made from the amino acid methionine, and the same pool serves the methylation of DNA and of the proteins that package it — the chemistry that determines which genes a cell reads. An enzyme consuming methyl groups at volume is competing with that, which is why inhibiting it has effects beyond the NAD+ arithmetic.

Where the Evidence Stands

The enzymology is well established: NNMT's reaction, its abundance in adipose and liver tissue, and its increase in obesity are documented in independent peer-reviewed work. That this compound inhibits the enzyme is also established, and it is used as a research inhibitor on that basis.

Functional work is at an early stage: reduced fat mass in diet-induced obese mice has been reported, and NNMT’s roles in liver and cancer biology are still being mapped. There are no human trials and no approved use, and it is supplied for laboratory research only. Its value is as a selective inhibitor of a defined enzyme, which is what makes the NAD+ and methyl-group questions testable.

Related Compounds

AICAR and SLU-PP-332 approach metabolic research by switching pathways on — one an energy sensor, the other a set of gene-regulating receptors. This compound does the opposite, removing an enzyme's activity, which usually produces a more interpretable result.

Its connection to NAD+ also places it beside NAD+ itself, supplied directly rather than protected from consumption — two quite different ways of asking the same question about supply.

Documentation and verification

Certificates of Analysis are published for tested products and batches. A certificate is a batch-specific record: it reports what was measured, by which method, on a stated date, against a batch identifier. It is evidence about the sample analysed at the time of analysis, and it is silent on attributes it does not list and on what has happened to the material since.

The batch number is the field that connects the document to a physical vial. Where the identifier on a vial does not appear on a certificate, the certificate does not describe that vial, however thorough the analysis behind it was. Batch records can be searched directly, and the guides below explain what each field establishes and what it does not.
