A Nuclear Receptor Agonist Studied for the Exercise Gene Programme

This compound belongs to the SLU-PP series of estrogen-related receptor agonists developed in the laboratory of Thomas Burris at Saint Louis University, and characterised in the peer-reviewed literature from 2023 onward. It is a small molecule rather than a peptide, and it acts on a class of protein quite different from the receptors most compounds in this catalogue engage.

Estrogen-related receptors — ERRα, ERRβ and ERRγ — are nuclear receptors, meaning they work inside the cell rather than on its surface, binding DNA directly and controlling which genes are read. Despite the name they are not activated by oestrogen. They were identified by their resemblance to the oestrogen receptor and were long classed as orphans, because no natural activating molecule was known.

What they govern is the machinery of aerobic metabolism: mitochondrial construction, fatty acid oxidation, and the genes muscle switches on when it adapts to endurance work. A molecule engaging all three subtypes activates that programme without the exercise that normally triggers it, which is the reason this class is described as an exercise mimetic and why it has attracted attention.

Why This Compound Is Studied

  • Deorphaning a nuclear receptor: ERRs had no known activating ligand for years, which made them very hard to study. A synthetic agonist is the tool that turns an orphan receptor into a subject that can be investigated experimentally.

  • Transcription rather than signalling: Acting on gene expression produces a slow, durable change in what a cell builds, unlike a surface receptor producing a rapid, temporary response. The two kinds of intervention behave differently and are studied for different questions.

  • Mitochondrial construction: The ERR programme includes the genes for building new mitochondria. This is a direct handle on a cell's aerobic capacity rather than on how it uses the capacity it already has.

  • Reported endurance findings: Mouse studies reported increased running capacity, reduced fat mass and improved glucose handling. Those findings are recent, from a small number of laboratories, and have not been extended to humans.

How It Works — In Simple Terms

Most receptors sit in the cell membrane and pass a message inward when something binds outside. Nuclear receptors do not. They live inside the cell, and when their activating molecule arrives they bind directly to DNA and change which genes are transcribed. The result is not a rapid response but a change in what the cell manufactures over hours and days.

The estrogen-related receptors control a coordinated set of these genes: the components of mitochondria, the enzymes of fatty acid oxidation, and much of the apparatus muscle uses for sustained aerobic work. This is the same programme endurance training switches on, which is why activating it pharmacologically is an interesting experiment.

The distinction from an energy-sensor compound is worth holding onto. Activating AMPK tells a cell to use what it has more aggressively. Activating ERRs tells it to build more of the machinery. One is an instruction about behaviour, the other about construction, and they operate on different timescales even when the measured endpoints overlap.

Where the Evidence Stands

This is genuinely new. The compound class was characterised in 2023 and the published work is mouse studies and cell-based assays from a small number of laboratories, principally the one that developed it. Reported findings include increased treadmill endurance, reduced fat mass in diet-induced obese mice and improved glucose tolerance.

No clinical trials have been registered or conducted and there is no human data; independent replication is limited because the work is recent. It is supplied for laboratory research only. The published characterisation is peer-reviewed academic work from 2023 onward, and the compound is the practical way to activate a receptor family that had no known ligand for years.

Related Compounds

AICAR is the established comparison: the same broad aim — activating the adaptations of aerobic exercise — reached through the cell's energy sensor rather than through gene transcription. Set side by side, the two make the difference between a signalling intervention and a transcriptional one unusually clear.

5-Amino-1MQ offers a third route, inhibiting an enzyme rather than activating a receptor, and is at a similarly early stage of investigation.

Documentation and verification

Certificates of Analysis are published for tested products and batches. A certificate is a batch-specific record: it reports what was measured, by which method, on a stated date, against a batch identifier. It is evidence about the sample analysed at the time of analysis, and it is silent on attributes it does not list and on what has happened to the material since.

The batch number is the field that connects the document to a physical vial. Where the identifier on a vial does not appear on a certificate, the certificate does not describe that vial, however thorough the analysis behind it was. Batch records can be searched directly, and the guides below explain what each field establishes and what it does not.
